Bile salts are a major component of bile, a fluid essential for the digestion of fats, absorption of fat-soluble vitamins, elimination of waste products, and formation of hormones. Bile and bile salts are made by the liver and stored in the gallbladder between meals. A deficiency in bile salts can cause poorly digested fats to pass through the digestive tract, causing flatulence, cramping, loose stools, and eventually an increased risk of gallstone formation.
Tauroursodeoxycholic acid, more commonly known as TUDCA, is a conjugated bile salt that may support gallbladder and liver health. Specifically, TUDCA has been shown to be a safe and effective support for patients with primary biliary cholangitis (PBC), a chronic autoimmune disease that causes bile to build up in the liver, and it may protect the liver from damage caused by liver cirrhosis due to long-term alcohol consumption. TUDCA has also been shown to slow the progression of metabolic dysfunction-associated fatty liver disease (MASLD) in mice fed a high-fat diet by reducing intestinal inflammation, improving intestinal barrier function, decreasing intestinal fat transport, and modulating the composition of the gut microbiota. Another study in mice also showed that TUDCA could potentially hinder the development of gallstones by reducing fat absorption in the intestinal tract.
While TUDCA’s role in liver and gallbladder health may seem more obvious, TUDCA also has a unique mechanism of action outside of biliary support. TUDCA has been shown to relieve endoplasmic reticulum (ER) stress in cells, thereby improving protein folding and cellular signaling. TUDCA’s ability to reduce ER stress extends its potential health benefits beyond liver health to a wide variety of chronic diseases that have demonstrated ER stress. Mechanistically, TUDCA also provides cellular protection by better regulating apoptosis.
One of the most important applications of TUDCA's cellular protective role is in the field of neurological health. Animal studies have shown that TUDCA serves as a neuroprotectant against acute injuries such as stroke, as well as chronic neurodegenerative diseases.
TUDCA also shows promise in reversing metabolic disorders caused by excess body fat and malnutrition. In fact, TUDCA has been found to significantly improve insulin sensitivity in liver tissue and muscle. In a study in aged mice, TUDCA reduced hyperinsulinemia and improved glucose regulation by enhancing the liver’s ability to break down insulin. Finally, TUDCA has also demonstrated protection against cell death in retinitis pigmentosa and defense against renal fibrosis, and even cardioprotective function by reducing scar tissue, cell death, and cardiac damage in mouse hearts subjected to increased stress.
